Chemotactic Cell Migration and Invasion Assay

The advent of permeable supports has provided a simple in vitro approach to measure chemoattractant-induced cell migration and invasion. The addition of a fluorescence blocking membrane improves the accuracy of the assay and allows the use of inverted fluorescence microscopy for detailed kinetic analysis. Fluorescence microscopy methods can accurately count the number of cells reaching the bottom of a membrane instead of relying on PMT-based measurements, which are more vulnerable to background fluorescence.

Step 1: Assay setup and analysis: FluoroBlok inserts, either untreated or containing biomatrix, are loaded with fluorescently labeled cells. Media containing chemoattractant is added to well reservoir below the insert. Migration of cells to the basolateral surface of the permeable support is characterized using kinetic fluorescence imaging and Agilent BioTek Gen5 image analysis tools.

Step 2: Results: This image-based method provides detailed profiles of cell migration over time and has considerable advantages over end-point assays and methods that rely on measuring total fluorescent signal.
