Macrophages and other phagocytes are specialized cells that consume and digest foreign matter or aberrant cells through phagocytosis. The pH-sensitive bioparticles or probes, including pHrodo-labeled Zymosan bioparticles, are a useful tool to study phagocytosis. These studies enable detailed quantitation of phagocytosis when coupled with kinetic live cell imaging and automated analysis.
Phagocytosis Assay Procedure
Step 1. Assay Setup: pH-sensitive probes that fluoresce once in the acidic environment of the phagolysosome are added to cell culture.
Step 2. Automated Kinetic Fluorescence Imaging: Phagocytic activity is revealed by an increase in bioparticle fluorescence over time.
Step 3. Automated Analysis: Agilent BioTek Gen5 microplate reader and imager software analysis tools automatically identify and quantitate changes in bioparticle fluorescence in real time.
