Microplate-based Signal Transduction Assays using Endogenously-expressed Drug TargetsTélécharger

Signal transduction refers to an intra-cellular response due to external stimuli. These responses are highly complex and involve multiple enzymes and their substrates in pathways that lead to myriad cellular responses, such as proliferation, migration, apoptosis, release of hormones, senescence among others. Protein kinases, along with epigenetic enzymes such as histone acetylases and deacetylases are involved in many signal transduction pathways and are of high interest as targets for intervention in disease, where their altered activity is the root cause of homeostatic imbalance. In this presentation, we will discuss biologically relevant assays, conducted in the cellular format where the enzyme is endogenously expressed. A number of automated microplate-based assays using both immortalized cell lines and human primary cells for the monitoring of the activity of endogenously expressed kinases and histone deacetylases will be described.
